I think it's a toss up (if you'll pardon the expression) between River and Amy.

I've come over all faint. Check out the covers (4 to choose from!) of this month's Doctor Who Magazine! Be warned there is a big spoiler/teaser. How long until season 6?

And the end of the season what? big deus ex machina
I bet River dies in the season opener. I don't want River to die again as she is an awesome character. But going by those covers (i'm going to get them all ), it's pretty much confirmed that one of them is leaving the show.
Teselecta: "Silence will fall when the question is asked."
Doctor: "And what is the question?"
Teselecta: "How much wood could a woodchuck chuck if a woodchuck could chuck wood?"

I don't think it's Amy as Karen Gillan (Amy) has filmed scenes for the episodes after the first two. Same with Arthur Darvill (Rory). I think River dies after we find out who she really is.
(But then again, thinking about it, that would already break continuity with her established death in "Forest Of The Dead" back in season 4/series 30 as technically, the River Song seen in season 5/series 31 and 6/32 is River Song before she meets the 10th Doctor in "Silence In The Library").

Very interesting twist that Moffats throwing in if thats true
As Easter draws nearer we're getting more information through about the explosive return of Doctor Who. There's some incredibly exciting adventures on the horizon and we've got the low down to share with you!
At one minute past midnight on Tuesday morning (5th April) we'll be bringing you an article in which Steven Moffat (Doctor Who's lead writer and Executive Producer) answers questions about the new series, plus we've plot details about the first two episodes.
